Actually, you can get this conversation even earlier. I had 495 points by the time I hit the central area of the Imp Fleet main ring; that was after getting the advanced class mission done, FYI. I saw Mako's conversation flag was active, so I talked to her; I then had 550 affection with the little scamp. (For what it's worth, I decided early on to do a Light Side Bounty Hunter.)

This is the first affection "mission" for Mako.

It's possible to get this as you wander into the cantina to do the mission for Minder Twenty
